An application server need not have any
JVMs per se...
However any EJB or servlet container is likely to have at least one JVM. There is no restriction on the number of VMs used by the app server.
It shouldn't matter, though. The number of JVMs should not affect the way you write your code, if you are otherwise following the EJB rules.
